The bodies of both the Hollywood director, 78, and his 68-year-old wife were reportedly discovered at their home in Los Angeles with what seemed to be knife wounds.
Reiner has directed classic films, including the likes of When Harry Met Sally, Stand By Me, The Princess Bride and This Is Spinal Tap, while Singer was an actress and photographer.
The Los Angeles Police Department has told ABC7 that the deaths are being investigated as homicides.
A family member is reportedly being questioned by police, but no arrests have been made.
A spokesperson for the family said: ‘It is with profound sorrow that we announce the tragic passing of Michele and Rob Reiner.

The couple met on the set of When Harry Met Sally and got married in 1989, the year it came out, and went onto have children Jake, Nick and Romy together.
Tracy, Reiner’s adopted daughter from his marriage to Penny Marshall, told NBC she had just seen her father the day before the incident for a family gathering.
‘I came from the greatest family ever,’ she said. ‘I don’t know what to say. I’m in shock.’
In a news conference, LAPD Deputy Chief Alan Hamilton has revealed the investigation is ongoing.
‘At this time, the Los Angeles Police Department is not seeking anyone as a suspect or as a person of interest,’ he said.

He confirmed that family members will be interviewed but insisted ‘no one has been detained’ and ‘no one is being questioned as a suspect’.
Hamilton said: ‘We’re going to try to speak to every family member that we can to get to the facts of this investigation.’
A neighbour told ABC7 that Billy Crystal – who starred in When Harry Met Sally – was seen looking ‘like he was about to cry’ as he visited the crime scene, while Larry David was also reportedly spotted visiting the scene.
LA Mayor Karen Bass said in a statement: ‘Rob Reiner’s contributions reverberate throughout American culture and society, and he has improved countless lives through his creative work and advocacy fighting for social and economic justice.’
She added that she was ‘heartbroken’ by the couple’s deaths, which she described as ‘a devastating loss for our city and our country’.
Reiner started his career as an actor, gaining attendance for his role in All In The Family, going onto appear in the likes of Sleepless in Seattle and The Wolf of Wall Street.

As a director, he became a real force in Hollywood, making his debut by the camera with the iconic mockumentary This Is Spinal Tap in 1984.
By the end of 80s, he’d added coming-of-age classic Stand By Me, fantasy comedy The Princess Bride, and rom-com When Harry Met Sally to his filmography.
‘Spinal Tap was satire, and I love satire, but that was something my father had done,’ he said to The Guardian in 2018, referencing his actor and director dad Carl Reiner.
‘And The Sure Thing was romantic comedy and he had done those. So Stand By Me was the first thing I did that was purely an extension of myself, and that meant a lot to me.’
He went continued to span genres, releasing Misery in 1990, and A Few Good Men in 1992.
The latter earned him a nomination for best picture Academy Awards, while he had four best director nominations at the Golden Globes.
He collaborated with Singer too, and their romance actually changed the plot of When Harry Met Sally.

He directed the likes of The Princess Pride (Picture: Century Fox/Kobal/Shutterstock) ‘It was going to be the two of them seeing each other after years, talking and then walking away from each other,’ he told CNN last year, before settling on the ending where they get married.
Reflecting on his marriage to Singer, he told Senior Planet: ‘We get along just great and enjoy each other’s company.’
In a post on X, Barack Obama said he and Michelle are ‘heartbroken by the tragic passing’ of the couple.
‘Rob’s achievements in film and television gave us some of our most cherished stories on screen,’ he continued. ‘But beneath all of the stories he produced was a deep belief in the goodness of people — and a lifelong commitment to putting that belief into action.’
